Tyrosine aminotransferase.

In plants, the pyridoxal-5′-phosphate (PLP)-dependent enzyme tyrosine aminotransferase (EC 2.6.15) is involved in the interconversion of tyrosine and 4-hydoxyphenylpyruvate, and phenylalanine and phenylpyruvate. Tyrosine and phenylalanine are amino acids that are ubiquitously involved in protein synthesis and are deemed proteogenic. In plants, these amino acids are also involved in reactions that are instrumental in the synthesis of compounds that are involved in plant secondary metabolism. Specifically, these two amino acids are catabolized in plants to provide precursor intermediates to facilitate the anabolism of phenylpropanoid compounds that are involved in plant responses to biotic and abiotic stresses. The model organism Arabidopsis thaliana provides an excellent tool to study aminotransferases because the genome of the organism has been sequenced and completely annotated. Recently, several tyrosine aminotransferases from Arabidopsis and several other plant species have been characterized using in vitro and in vivo biochemical analyses. The results from these studies show that, unlike the bacterial enzyme, which is primarily involved in the synthesis of tyrosine and phenylalanine to facilitate protein synthesis, the major role of plant the plant enzyme is in the biosynthesis of compounds that are involved in secondary metabolism facilitated through the catabolism of tyrosine and phenylalanine. Recent studies have confirmed that tyrosine aminotransferase is involved in many aspects of secondary metabolism; these include the biosynthesis of compounds such as rosmarinic acid in Salvia miltiorrhiza and tocopherol in Arabidopsis thaliana. The goal of this chapter is to present a synthesis of the recent work on tyrosine aminotransferases in plants by highlighting the kinetic and physical properties of the enzyme in addition to its overarching role(s) in plants.
